    Surrogate Signature Development
  • Developed a surrogate signature for a target gene and validated it based on expression correlation through expression correlation analysis utilizing Fischer’s test, OS and DFS survival analysis, hazard ratio assessment, response to immune checkpoint therapy, and hallmark signature correlation parameters (single-cell and bulk RNA seq data).
  • Surrogate Signature validation and elucidation of immune exclusion mechanisms through Spatial Transcriptomics Analysis
  • Analyzed target gene's correlation with immune exclusion metric (CD8 density ratio stroma/epithelial).
  • Identified the primary cancer-associated fibroblast (CAF) subtype expressing the target gene by examining its association with the surrogate signature and the four main subtypes of CAFs.
  • Determination of target gene’s Cellular Composition and Mutation-Driven Expression Variability
  • Determined mutation markers associated with both high and low target gene expression by hypothesis-based testing (CCLE & TCGA data). Investigated the impact of joint mutations on target gene's expression in NSCLC at the bulk tumor level (TCGA data).
  • Executed pan-cancer analysis data to elucidate the cellular composition linked to the target gene and its receptor expression. Generating insightful visualizations through volcano plots, heatmaps, and scatterplots.
